WINSTON-SALEM, N.C. July 31, 2013 Could harvesting stem cells for therapy one day be as simple as asking patients for a urine sample? Researchers at Wake Forest Baptist Medical Center's Institute for Regenerative Medicine and colleagues have identified stem cells in urine that can be directed to become multiple cell types.

"These cells can be obtained through a simple, non-invasive low-cost approach that avoids surgical procedures," said Yuanyuan Zhang, M.D., Ph.D., assistant professor of regenerative medicine and senior researcher on the project.

Reporting online in the journal Stem Cells, the team successfully directed stem cells from urine to become bladder-type cells, such as smooth muscle and urothelial, the cells that line the bladder. But the urine-derived cells could also form bone, cartilage, fat, skeletal muscle, nerve, and endothelial cells, which line blood vessels. The multipotency of the cells suggests their use in a variety of therapies.

"These stem cells represent virtually a limitless supply of autologous cells for treating not only urology-related conditions such as kidney disease, urinary incontinence and erectile dysfunction, but could be used in other fields as well," said Zhang. "They could also potentially be used to engineer replacement bladders, urine tubes and other urologic organs."

Being able to use a patient's own stem cells for therapy is considered advantageous because they do not induce immune responses or rejection. However, because tissue-specific cells are a very small subpopulation of cells, they can be difficult to isolate from organs and tissues.

Zhang's team first identified the cells, which are a small subset of the many cells found in urine, in 2006. The current research builds on earlier studies by confirming the multipotency of the cells. In addition, the research found that unlike iPS cells or embryonic stem cells, the urine derived-stem cells do not form tumors when implanted in the body, indicating they may be safe for use in patients.

The research involved obtaining urine samples from 17 healthy individuals ranging in age from five to 75 years. Isolating the cells from urine involves minimal processing, according to the authors. Next, they evaluated the cells' ability to become multiple cell types.

Importantly, the cells differentiated into the three tissue layers (endoderm, ectoderm and mesoderm) that are a hallmark of true stem cells and also differentiated into the specific cell types mentioned earlier.

Next, the researchers placed cells that had been differentiated into smooth muscle and urothelial cells onto scaffolds made of pig intestine. When implanted in mice for one month, the cells formed multi-layer, tissue-like structures.

The urine-derived stem cells have markers of mesenchymal cells, which are adult stem cells from connective tissue such as bone marrow. They also have markers for pericytes, a subset of mesenchymal cells found in small blood vessels.

Where do the cells come from? Researchers suspect that the cells originate from the upper urinary tract, including the kidney. Female study participants who had received kidney transplants from male donors were found to have the y chromosome in their urine-derived stem cells, suggesting the kidney as the source of the cells.

"Identifying the origins of the cells will lead to a better understanding of the biology of this multipotent population of mesenchymal cells within the urinary tract system," said Zhang.

'Housewives' stars free on bond, travel restricted

NEWARK, N.J. (AP) ? A star of "The Real Housewives of New Jersey" and her husband were each released on $500,000 bond after making initial court appearances Tuesday on federal fraud charges.

Teresa Giudice and her husband, Giuseppe "Joe" Giudice, were ordered to surrender their passports and restrict their travels to New Jersey and New York. Joe Giudice, an Italian citizen, could be deported if convicted.

The Giudices, who rose to celebrity status by showcasing their lavish lifestyle on the Bravo reality TV show, were indicted Monday on bank and bankruptcy fraud charges, among other counts.

Joe Giudice is also charged with failing to file tax returns from 2004 to 2008, when he is alleged to have earned nearly $1 million.

The couple is accused of exaggerating their income while applying for loans before their show debuted in 2009, then hiding their fortunes in a bankruptcy filing after their first season aired.

The couple did not enter pleas during the brief appearance in a courtroom packed with media and spectators. Their arraignment was set for Aug. 14. Their attorneys said both will plead not guilty.

"The investigation went on for a pretty long time and we are confident we have enough evidence to convict the defendants beyond a reasonable doubt," U.S. Attorney Paul Fishman said after the proceeding.

Both face hundreds of years in prison and millions of dollars in fines if convicted, but federal sentencing guidelines would greatly reduce the penalties, Fishman said.

Teresa Giudice's attorney, Henry Klingeman, said he believes the government is prosecuting the couple because of their celebrity.

The couple filed for bankruptcy in 2009, claiming they owed $11 million, including $2.2 million in mortgages, $13,000 to Neiman Marcus and Nordstrom, and nearly $12,000 to a fertility clinic.

Joe Giudice also is facing charges in Passaic County that he used his brother's identity to obtain a driver's license, presenting his marriage and birth certificates. He rejected a plea deal in April through his lawyer, Miles Feinstein.

"In Passaic County, where I represent him, I feel that because of his celebrity status he's being treated differently than others are for the same offense," said Feinstein, who did not comment on the federal case.

Teresa Giudice has parlayed her fame into cookbooks, a line of ready-made bellinis and "Skinny Italian," a specialty food line.

On the show, she is known for her expensive tastes and combative relationship with her brother and sister-in-law. But Klingeman noted that the show helps support the couple and their four children.

"While it's called reality TV," Klingeman said, "I'm not sure everything you see on the show is real."

Adventures with Jaap: Home Sweet Home

Jaap and I spent the first two weeks of May visiting family and friends in NY. It's always good to come home and this time we got to celebrate the pregnancy (complete with a surprise baby shower!) with family and friends.?

No trip to NY would be complete without an evening with Shannon, my best friend, and our NY buddies. After a night out on the Lower East Side we enjoyed a delicious breakfast at Floridita. If you want the best coffee in NYC, go to Floridita and order a cafe con leche. It's up on the West side on 125th St and Riverside Drive.?

What was planned as a casual lunch with my aunts turned out to be a surprise baby shower complete with Muppets! Thanks to my amazing sister and mother for organizing the day. It was such a great surprise to see our closest famiy and friends there. What a day. We got so much great stuff for the baby and Jaap was able to celebrate in Muppet style. We felt very loved. Now we just wait for the little Jansen-Pardo to arrive!

Loeb's Third Point opens stake in CF Industries: letter

NEW YORK (Reuters) - Daniel Loeb's hedge fund Third Point initiated a new position in nitrogen fertilizer manufacturer CF industries during the second quarter, according to a quarterly investor note.

"CF currently trades at an unwarranted discount to fertilizer and commodity chemical peers," Loeb said in the letter, dated July 29. "We believe its structural cash flow generation strength is misunderstood and that management should deliver a much larger dividend to its shareholders."

Loeb said that CF's "access to low-cost North American natural gas - the primary input in nitrogen fertilizer production - gives the company a structural, sustainable margin capture relative to global peers with higher input costs."

Top Credit Improvement Tips To Get You Going | - JGVFinance

Have you been suffering from poor credit for years? Many people have credit scores that are getting worse. Fortunately, a number of methods for improving such ratings do exist, and you can begin using them right away by reading these helpful hints.

Try to keep a balance of less than 50% of your available credit on all of your cards. When balances are over 50%, your credit rating goes down significantly, so try to either spread out your debt or, ideally, pay off your credit cards.

Work with the companies to whom you owe money to get your debt back under control. This will help you stabilize your situation and start working towards a better financial situation. Contact your credit card company and request to change your scheduled due date or interest rate.

Credit Counseling

Consumers should carefully research credit counseling agencies before choosing one with which to work. You will find some counselors that truly want to help you fix your credit situation, while others may have different motives. Some are not legitimate. Intelligent customers will make sure to determine the legitimacy of a credit counseling agency before acquiring their services.

Stay cautious and aware of scams online that can lead you to even bigger problems. A common scam involves teaching you how to make a completely new, albeit fraudulent, credit file. This is illegal and you will eventually be caught. The end result of getting caught during the crime could be expensive legal fees and a possibility of time behind bars.

Call and request lower limits on your cards from the credit card companies. Doing this keeps you from overtaxing yourself. It also shows the lending company that you are responsible.

You should look over all negative reports thoroughly when attempting to fix your credit. Errors are not infrequent in credit reports. If you can prove the credit bureau is in the wrong, they should correct the reported error or remove it entirely.

Find and challenge errors on your credit report using the dispute process offered by each credit reporting agency. Draft a letter to reporting agencies disputing negative entries and also submit any available documentation. Send your dispute package with a return receipt request so that you have proof that it was received by the agency.

It is crucial that you review credit card bills on a monthly basis to check for errors. If this is the case, you need to call the company right away to avoid them from reporting it to credit reporting agencies.

Any time you establish any payment plan with any creditor, make sure you get it in writing. This is a great way to have documentation of the plan if the creditor changes their mind or the company ownership gets changed. As soon as you get it paid off, have that in writing so you are able to inform the credit reporting agencies.

You may want to argue against the reports, but potential lenders will not take your statements into consideration. The action of making a statement about your negative history will work against you as it highlight your mistakes rather than downplaying them.

Collection Agencies

Some of the worst stress you experience with bad credit is caused by debt collection agencies. A consumer has the legal option of using cease and desist letters to dissuade collection agencies, but it is vital to keep in mind that C&D letters only stop harassment. This doesn?t let the customer off the hook for the debt, it merely stops the threatening calls.

Be careful of paying for a service or a lawyer that advertises quick or instant credit score improvement, many of them are dishonest. A lot of people are having difficulty with their credit, and there are lawyers that try to exploit these people with illegal and ineffective credit restoration services. Do your homework and check out any attorney before you hire one to help with credit improvement.

Part of a nasty credit crunch is having multiple debts that you do not have the money to pay. Try to make sure that you find a little bit of money in your budget for all the creditors you owe payments to. Making the minimum payments will, at least, avoid ending up with collection agencies.

You need a plan and schedule when it comes to paying off your debt. These accounts will still appear on a credit report, but they will be earmarked as paid.

If a poor credit rating has left you feeling discouraged, turn your situation around by applying the guidelines you have just read. They can stop your credit from falling any further, and get you back on the road to recovery.

Hunter Mahan chooses his daughter's birth over golf

Hunter Mahan was leading the RBC Canadian Open golf tournament on Saturday. But then his wife went into labor. So, Hunter Mahan withdrew from the tournament.

Adios! Hunter Mahan waves after sinking a birdie putt on the 18th hole during the second round at the Canadian Open golf tournament in Oakville, Ontario. Mahan was leading the tournament on Saturday July 27, when he withdrew to attend the birth of his first child.

Players are expected to call penalties on themselves, even if no one is looking.

That's why it's fitting that Hunter Mahan ? who was leading the RBC Canadian Open after two rounds ?? withdrew to attend the birth of his daughter. Mahan did the right thing ? for himself, for golf, and for fathers everywhere.

That's not to say that it was an easy decision. Mahan hasn't won a PGA tournament this year. He finished in the top 10 in two recent big tournaments ? the US Open and the British Open Championship. Going into Saturday's third round, he was leading by two shots. And if he had won, Mahan would have pocketed a $1,008,000 winner's check.

How many new dad's would walk away from a $1 million pay day?

The baby wasn't due for a few weeks. But then his phone rang Saturday morning, while he was warming up on the practice range. His wife was in labor, her water had broken. He didn't hesitate. He politely left the tournament.

?I received exciting news a short time ago that my wife Kandi has gone into labor with our first child,? said Mahan. ?As a result, I have withdrawn from the RBC Canadian Open to return to Dallas. I would like to extend my very sincere gratitude and appreciation to RBC and the RBC Canadian Open.

"Kandi and I are thrilled about this addition to the Mahan family and we look forward to returning to the RBC Canadian Open in the coming years.?

Mahan is not the first golfer to face the challenge of fatherhood vs. work. As Yahoo Sports writer Jay Busbee wrote: "Back in 1999, Phil Mickelson famously wore a beeper during the U.S. Open. His daughter was born the day after Mickelson lost to Payne Stewart. More recently, Ross Fisher vowed to walk off the course at the 2009 British Open no matter what if he heard his wife was going into labor; he was just one stroke behind leader Tom Watson at the time."

And last fall, Chicago Bears cornerback Charles Tillman caused a stir when he suggested that the birth of his child would take precedent over showing up for his job. It sparked conversations on talk radio and in social media about work and family priorities. Tillman said:

"[Football will] always be second or third in my life. That was a great lesson learned, to teach me that family ? when I?m done playing football ? my family will always be there for me.?

As The Christian Science Monitor reported at the time, "Tillman's announcement drew criticism from some sports columnists and fans. Sport radio talk show starkly framed the question: "If you only work 16 days a year, should you miss one for the birth of your child?"

Tillman's daughter was eventually born on a Monday, saving the NFL player and fans from any further controversy.

But Mahan's daughter Zoe arrived on a work day. As a father, it's unlikely Mahan will regret the decision to leave the golf tournament. And there are probably more than a few dads ? who missed the birth of their first child ? who would now gladly give a $1 million to be there.

Microsoft's Windows NT turns 20

OSNews writes, Windows NT turned 20 years old today. From a 1998 story about NT, written by Mark Russinovich, "I'll take you on a short tour of NT's lineage, which leads back to Digital and its VMS OS. Most of NT's lead developers, including VMS's chief architect, came from Digital, and their background heavily influenced NT's development. After I talk about NT's roots, I'll discuss the more-than-coincidental similarities between NT and VMS, and how Digital reacted to NT's release."

Another Possible Bright Supernova Discovered In Spiral Galaxy M74

I love this galaxy. Not only does M74 display a near perfect spiral form but if this latest supernova is the third to ?go boom? in the galaxy in just 11 years. The new object, designated PSN J01364816+1545310, was discovered blazing near 12.4 magnitude by the?Lick Observatory Supernova Search?at Lick Observatory near San Jose, Calif. ?PSN? stands for ?possible supernova? and the long string of numbers give the object?s position in the sky using the celestial equivalents of latitude and longitude.

M74 is a classic spiral galaxy with arms that appear to unwind from a bright, star-packed nucleus. Located 32 million light years away in the constellation Pisces, M74 contains about 100 billion stars. The spiral arms are dotted with dense star clusters and pink clouds of fluorescing hydrogen gas. The Lick search uses a fully robotic or automated 30-inch (76 cm) telescope dedicated to scanning the skies for new supernovae. It nailed M74?s latest exploding star on July 25. Two previous supernovae flared in the galaxy -?SN 2002ap?and?SN 2003gd?- and rose to 12th and 13th magnitude respectively before fading away into obscurity.

Three?s the charm as they say. A team of astronomers using a spectrograph at the Faulkes Telescope South at Siding Spring, Australia teased apart the supernova?s light and now know exactly what blew up. It appears our newcomer was originally a supergiant star at least 8 times as massive as the sun.?After a relatively brief lifetime measured in the millions of years, the supergiant gobbled up the last of its fuel. With the gas gauge on ?empty? and no new energy being produced in the core to hold back ?the force of gravity, the star imploded, sending a shockwave rocketing back in the opposite direction that tore it to bits.

When a massive star runs out of nuclear fuel in its core, the energy that has prevented the force of gravity from crushing the star is gone. Gravity now finally wins and collapses the star which then rebounds in a huge explosion. Credit: ESO

Gravity finally gains the upper hand causing the star to implode. A rebounding shock wave blows it to bits. Sometime a city-sized, dense stellar remnant called a neutron star remains after the blast. Credit: ESO

Called a Type II supernova explosion, the blast hurtles star stuff into space at up to 45,000 miles per second (70,000 km/sec). More amazing, a powerful supernova explosion can release as much energy as the sun during its entire 10 billion year lifetime. No wonder even small telescopes can spot these cataclysmic events from millions of light years away!

The galaxy M74, the 74th entry in 18th century astronomer Charles Messier's catalog, is found about 1.5 degrees east-northeast of the star Eta Piscium just to the right of the small constellation Aries the Ram. The map shows the sky around 1 a.m. tomorrow morning facing east. Stellarium

As additional photos and measurements come in, ?amateur astronomers with 8-inch and larger telescopes will have no problem spying the supernova once the last quarter moon departs the vicinity. It?s located 93? (1.5?) ?east and 135? (more than 2?) southeast of the galaxy?s core. The map and photo will help you track it down.

This map measures only about 1/2-degree wide and shows the galaxy up close with the supernova marked SN. Selected star magnitudes from the AAVSO are shown to help you navigate to the object as well as estimate its brightness. North is up, west to the right. Map created with Chris Marriott's SkyMap software

While M74 is relatively bright and appears spectacular in long-exposure photos, it looks like a large, dim featureless glow in smaller telescopes. Be patient and take your time to ?star hop? to the supernova using the more detailed map. Matter of fact, you may want to wait until Tuesday morning or later to look. That?s when the waning moon will finally depart the area. Let?s hope our new guest remains bright.

Good luck meeting the latest star to mark the end of its life with the biggest blowout of all. For more information and photos, stop by Dave Bishop?s Latest Supernovae site.
